Phelps's appearance on the Sept. 13 edition of the iconic sketch comedy series marks his acting debut, the network said.
Recording artist Lil Wayne will be the show's musical guest that night.
NBC said the "SNL" season premiere will kick off four live shows in a row. Seven all-new episodes are scheduled to air Saturdays prior to the U.S. presidential election.
Phelps went 8-for-8 in Beijing and broke Mark Spitz's single-Games record for most gold medals won.
In addition, Phelps became the most decorated Olympian in history, winning 14 career gold medals.
